[SHORT REPORTS]Mechanism Involved in the Different Responsiveness to Thyroid Hormone between Monolayer and Spheroid Cultures of Rat Hepatocytes

We demonstrated in a previous study that the responsiveness to thyroid hormone was well preserved in the hepatocytes of spheroid cultures but not in monolayer cultures. It was speculated that the differences could be due to changes in the retinoid X receptor α (RXRα) or the c-Jun expressions which have been reported to affect the action of the thyroid hormone. We undertook this study to evaluate the expression of the RXRα and c-jun mRNA in the cultured hepatocytes from rats. A slight but significant increase in RXRα was observed in the spheroid cultures, although there was no clear difference in the expression of the c-jum mRNA. These results suggest that the increased expression of RXRα mRNA may be responsible for maintaining the 3,5,3'-triiodo-Lthyronine (T_3) responsiveness in spheroid cultures.
