When plant protoplasts have been isolated by enzymic method, usually two principal methods have been used (8). In the sequential method, leaves are cut into small pieces after the lower epidermis is removed and the pieces are incubated in pectinase. This procedure causes the maceration of tissues and the release of mesophyll cells. After that, the cells are suspended in a cellulase solution to digest the cell walls and release the protoplasts. In one-step methods, a mixture of pectinase and cellulase is used that not only macerates the tissues but also releases protoplasts by digesting cell walls. Meicelase P-1 is usually classified into cellulase (2). However, this enzyme have pectinase activity, too. Therefore, this enzyme releases protoplasts from some plant leaves without addition of pectinase. This paper described the action of Meiselase P-1 for the isolation of protoplasts from rice, lettuce, sugarcane and coffee leaves.
