Biological effects of bisphenol-A assessed by induction of apoptosis in vitro

We examined the biological effects of bisphenol-A (BPA), 17 beta-estradiol (E_2) and methyl methacrylate (MMA) on induction of apoptosis in MCF-7 cells in vitro. We used annexin V-FITC staining, DNA laddering analysis and the activities of caspase-3 and caspase-6 for evaluating induction of apoptosis, and E-screen for estrogenicity. The three chemicals did not induce apoptosis at day 1. At day 3 BPA induced apoptosis at a rate of 5% at a concentration between 1 × 10^<-8> M and 1 × 10^<-5> M, MMA induced it between 1 × 10^<-8> M and 1 × 10^<-4> M. E_2 induced apoptosis at a rate of 10% at a concentration between 1 × 10^<-12> M and 1 × 10^<-6> M at day 3. The three chemicals showed an increase in induction of apoptosis and necrosis in the same concentration range at day 6, when compared with day 3. BPA seemed to induce excessive necrosis by cytotoxic effect at 1 × 10^<-4> M at day 6. Although BPA and E_2 yielded estrogenicity with E-screen, MMA did not. These results indicate that BPA induces apoptosis and estrogenic activity in MCF-7 cells in a similar range of concentrations. Further study on the physiological aspects of apoptosis induction by BPA is necessary to elucidate its biological effect on cells.
