Pit-1 (GHF-1) is one fo the POU-domain transcription factors. It activates the transcription of growth hormone (GH) and Prolactin (PRL) genes and plays a critical role in the normal development of the somatotroph. Iactroph and thyrotroph. To investigate the relationship between the levels of PRL and Pit-1 mRNAs under estrogen administration, estradiol dipropionate or estrogen pellets were administered to male Wister rats by either single intra-muscular injection or subcutaneous implantation using silastic tubes. Messenger RNAs for PRL and Pit-1 in the anterior pituitary were determined by Northern blot. Estrogen administration resulted in a significant increase in PRL mRNA. However, Pit-1 mRNA was not altered. The results suggest that the increase in Pit-1 mRNA is not required for the activation of the PRL gene expressionby estrgen.
