Light microscopic observatiaons showed that silk materials, fibroin and sericin (which were deposited in the lumen of the silk gland during the 4th instar) are dissolved during the early and middle period of the molting stage. Furthermore, the production of these silk materials in the gland cell is stopped at the early period of the 4th molting stage. At the same time remarkable structural changes occur including the appearance of numerous vacuoles in the cytoplasm and the reduction of cytoplasmic basophila. Late in the molt the gland cell regains its basophila and the vacuoles disappear.Striking ultrastructural changes also occur during the 4th molt. The amount of granular endoplasmic reticulum in the gland cell decreases. Both the formation within Golgi vacuoles and the secretion into the lumen of the fibroin also decreases conspicuously during the molting stage, but the secretion of small amount of fibroin continues until ecdysis.Numerious vacuoles and intercellular spaces appear during the molt, and these are interpreted as necessary for the intracellular transport of fluids through the cell layer. The new tracheal system develops in the gland cell as tracheoblasts invade the intercellular spaces. Lysosome-like bodies aggregate in various regions in the gland cell at ecdysis. The relationship between these ultrastructural changes and the fluid transport in the gland cell during molting stage is discussed.
