投影X線顕微鏡によるウニの受精卵の内部構造の観察

概要

The biological application of projection x-ray microscopy has been continued using the imaging x-rays at the wavelength range of 0.5-1 nm in order to ascertain whether the inner structures of the mitotic apparatus in sea-urchin eggs are detectable or not. As a result, it is found that spindle fibers, astral microtubules, chromosomes, and centrosomes of the mitotic apparatus are well recognized in the sample prepared by critical point drying of stabilized sea-urchin eggs using MEG buffer, though the structures of these mitotic apparatus are obscured to some extent by superposition of surface structures of the eggs, while no structures are recognized in the case of the natural drying. These structures are seen three dimentionally with stereoscopic pair. Image processing is successfully tried to improve the image resolution and contrast.
