HeLa cell lines that constitutively expressed the fusion (F) and/or hemagglutinin-neuraminidase (HN) glycopriteins of human parainfluenza virus type 4B (HPIV-4B) were established, and they were used to analyze the roles of these glycoproteins in virus-induced cell fusion and to compare the properties of the glycoproteins of HPIV-4A. Though co-cultivation of HeLa cells expressing F protein of HPIV-4A (4A-F-HeLa) and HeLa cells expressing both the F and HN proteins of HPIV-4A (4A-F+HN-HeLa) generated large polykalyocytes, no syncytium formation was found in the co-cultivation of 4B-F-HeLa and 4B-F+HN-HeLa. However, cell fusion was found both in the co-cultivation of 4A-F-HeLa and 4B-F+HN-HeLa and in that of 4B-F-HeLa and 4A-F+HN-HeLa, indicating that expressed glycoproteins of HPIV-4B had some functions in cell fusion. Monoclonal antibodies against HPIV-4 glycoproteins had no effect on the cell fusion, but polyclonal antibodies inhibited the cell fusion.
