The following volumetric method for the determination of cellulose and hemicellulose is accurate and can be carried out much more rapidly than the gravimetric method. This method is based on the fact that cellulose can be quantitatively oxidized with potassium dichromate in sulfuric acid solution. C_6H_<10>O_5+6O_2=6CO_2+5H_2O (1) cellulose determination in alkalicellulose Alkalicellulose is neutlized by sulfulic acid solution, the cellulose obtained in this procedure by filtration through Gooch crucible, and dissolved in 30cc. of 80% sulfuric acid, and diluted to 100cc. A 10cc. sample is pipetted in a 500cc. pyrex beaker, to which 10cc. standard dichromate solution and approximately 50cc. of 72% sulfuric acid solution are added. The solution is boiled gently for about 5 min, cooled in water, and the excess dichromate is titrated with ferrous ammonium sulfate solution, using barium diphenylaminesulfonate as an indicator. (2) hemicellulose determination in alkalicellulose Alkalicellulose is treated with 8% sodium hydroxide solution for 30 min. in 100cc, mess-flask, and then filtered through a glass filter (1 G 3) or a Gooch crucible. A 25cc. portion of this solution is pipetted in a 500cc. beaker and a 5cc. of the standard dichromate solution and 50cc. of 90% sulfuric acid solution are added. The content of the beaker is boiled about 5 min., cooled in water, and then titrated as the same as the cellulose determination.
