The origin of delta bilirubin (δBr) was studied. Analbuminemic rats (NAR), Sprague-Dawley (SD) rats and guinea pigs were subjected to bile duct ligation (BDL). Serum bilirubin was fractionated by high performance liquid chromatography (HPLC) at timed intervals. Six of the NAR were injected with rat albumin (2 μmol) a day after BDL. For 3 days after BDL serum bilirubin increased to 156±34μmol/l and decreased thereafter in the SD rats. However, the increase was small (28±6μmol/l) in the NAR (n=4) and no δBr appeared. δBr appeared and increased in the NAR treated with albumin. δBr also appeared after BDL in the serum of the guinea pigs, who excreted more than 90% of bilirubin as bilirubin monoglucuronide (BMG). Bilirubin diglucuronide (BDG), BMG or unconjugated bilirubin (UCB) was incubated with bovine serum albumin (BSA) at 37℃ in the dark under argon atmosphere, and δBr formation was observed. Concentrations of BDG, BMG, UCB and BSA were adjusted to 150μmol/l. About 12.3±1.2%, 2.8±0.2% and 0.3±0.1% of the initial bilirubin amount were converted to δBr from BDG, BMG and UCB, respectively, after 6 hours of incubation with BSA. From these results, it is concluded that (1) BDG is the main source of δBr formation in vitro, although δBr can be formed from BMG in guinea pigs, and (2) albumin is necessary for δBr formation.
