In this study using female mice of different strains (7-9 weeks of age) primary mixed lymphocyte reactions (MLR) revealed that the Lyt-2^+ and L3T4^+ T cell subsets from C57BL/6 (B6) mice were capable of responding, exclusively to class I H-2 [B6-C-H-2^<bm1>(bm1)]-and to class II H-2[B6-C-H-2^<bm12>(bm12)]-alloantigens, respectively. Anti-bm12 MLR was not affected by the i.v. injection of bm12 spleen cells to the recipient B6 mice. In contrast, a single i.v. administration of bm1 spleen cells to B6 mice resulted in the abrogation of the capacity of the recipient B6 lymph node cells to give anti-bm1 MLR. In order to investigate cellular requirements for such tolerance induction, the allogeneic bml spleen cells were fractionated into various lymphoid cell fractions including antigen-presenting cell (APC)-depleted (T and B cell) and APC-enriched fractions, and the experiment for presensitization using the fractions was made. The results demonstrated that the recipient B6 anti-bm1 Lyt-2^+ helper T cell tolerance was inducible with either type of the above lymphoid cell fractions. However, the administration of soluble bm1 alloantigens to the recipient B6 mice failed to induce anti-bm1 tolerance. And further, when cells of the spleen from (bm1×bm12) F_1 mice were injected i. v. to the recipient B6 mice, those B6 mice exhibited anti-bm1 tolerance along with the positive response against bm12 alloantigens. These results indicated the distinctiveness of class I H-2 alloantigen-reactive Lyt2^+ helper/proliferative T cell subset in the aspect of tolerance induction, also giving valuable information for discussion in relation to the different effects of donor-specific transfusion on the regulation of host's immune response to alloantigens.
