Goodpasture (GP) antigens, protein components reactive with human autoantibodies against glomerular basement membrane (GBM), were identified in human alveolar basement membrane (ABM), by an enzyme-linked immunosorbent assay (ELISA), Western blotting and immunoprecipitation. All the six anti-GBM antisera studied, three obtained from patients with glomerulonephritis and pulmonary hemorrhages (i. e. GP syndrome) and the other three from patients with glomerulonephritis alone, distinctively reacted with the ABM prepared from normal human lungs by a detergent method and solubilized with bacterial collagenase. Very cationic 22-28 kDa and 40-48 kDa components were detected by the blot analysis in combination with either SDS-polyacrylamide gel electrophoresis (SDS-PAGE) or two-dimensional gel electrophoresis. These proteins showed some homology with the GP antigens in human GBM, as far as monomer-dimer composition and charge distribution are concerned. Inhibition ELISA also revealed that the binding of anti-GBM antisera to collagenase-digested (CD) GBM decreased when the antisera were preincubated with CD-ABM, which suggests that the anti-GBM antisera recognized the same epitope (s) on the GBM and the ABM. Further, the heterogeneity of GP antigens in human ABM was demonstrated by SDS-PAGE/blotting ; monomeric antigens were absent or at minimal levels in CD-ABM of the three out of 10 normal individuals examined, although proteins were detectable by staining in the monomer region of SDS-PAGE gels. In immunoprecipitation, anti-GBM antisera from patients with pulmonary hemorrhages and those from patients without the hemorrhages showed different reactivities with CD-ABM ; the former antisera precipitated both monomeric and dimeric components, but the latter did not precipitate any proteins. These results indicated that the GP antigens in human ABM had some homology with those in human GBM. The observations of the variation in monomer-dimer composition of ABM, and the difference in binding of anti-GBM antisera to ABM may explain why only some and not all the patients with anti-GBM nephritis have lung involvement.
