<原著>糖尿病性消化管機能異常における自律神経組織およびvasoactive intestinal peptide含有神経の形態的研究

In order to investigate the relationships between the intestinal autonomic nervous system and vasoactive intestinal peptide (VIP) in intestinal disorders, diabetes mellitus was induced in 46 Wistar rats by intravenous administration of 30 or 40 mg/kg streptozotocin (SZ). The controls were 43 age-matched rats. The duodenum, jejunum, ileum, cecum, colon and rectum were examined by using HE and silver-impregnation, electron microscopic preparations, the light- and the electron-microscopic observations of immunoreaction to VIP antibody, and the quantification of VIP in the ileum and colon in the experimental and the control animals. The morphological appearances of the autonomic nervous tissue in diabetes mellitus (DM) were similar to controls throughout the intestines except for the colon and cecum, which grossly showed significant dilatation and the thinned walls. The results can be summarized as follows. 1. Electron microscopic changes in most of the submucous and myenteric plexuses examined were nonspecific, although they were more predominant in the cecum and colon. These changes included the presence of myelin figures, filamentous inclusions, membranous structures, aggregates of microtubules in the axons, and electron-dense materials in the neurons. 2. Immunostaining for VIP in the submucous and myenteric plexuses revealed marked varicosity of the axons and coarse disordered fiber networks. Immunoelectron microscopy revealed that the number of reaction-positive fibers was significantly increased in proportion to the total number of axons. 3. Quantitative analysis of VIP revealed the increase in the ileum and colon. 4. Synaptic contact between VIP-positive axons and reaction-negative axons was found in the control animals. This has not been described yet in the references. It is clear from these data that VIP in the axons of autonomic nervous system is closely related to disorders of the digestive system in the cases of diabetes mellitus.
