By using enzyme-linked immunosorbent assay (ELISA), the antibody levels against the crude antigens of Campylobacter jejuni (C. jejuni) were studied in the sera from 36 patients with C. jejuni enteritis. In the period between the second and the eighth week of illness, the antibodies were detected in the sera of all the patients but a patient receiving immunosuppressive agents for an underlining disease. For the present study, isolation and purification of an antigen of C. jejuni were performed : A strain (Hisaichi strain), isolated from a patient with C. jejuni enteritis, was cultured on blood agar plates. Soluble crude antigen(s) was extracted from the homogenized bacteria, and was chromatographed through a Sepharose CL-4B column followed by DEAE-Sephadex A-25 column. The purified antigen (C. j. antigen) showed almost one band in the SDS polyacrylamide gel electrophoresis, which suggests its homogeneity. Sera from 28 of the 36 (78%) patients were proved to be positive by ELISA on the purified C. j. antigen. None of the sera from 15 non-C. jejuni enteritis reacted with the purified C. j. antigen. These data indicated that the purified C. j. antigen, a main antigen in the immune response to C. jejuni in the majority of the patients, was useful for ELISA coating to diagnose C. jejuni enteritis and to clarify the antigen-antibody system in C. jejuni infections. Antibody against purified Escherichia coli heat-labile enterotoxin (LT) was demonstrated in 17 of the 35 (49%) sera of patients with C. jejuni. The antibody titers, however, were not so high as those in the sera from patients with cholera, which suggests that some strains of C. jejuni can produce a toxin or toxin-like substance, immunologically related to LT.
