<原著>酵素抗体法による乳癌を中心とした乳腺組織のEstrogen Receptorの細胞組織化学的研究

We made cytohistochemical studies on the estrogen receptor (ER) in breast carcinoma and other breast diseases by the immunoperoxidase method to clarify the mechanism of the carcinogenesis and the adaptative standard of the endocrine treatment of breast cancer. Ten percent formalin is the better fixative to be used for fixation in the immunoperoxidase method, and it is applicable to general pathohistological uses. In the benign breast diseases, on the normal mammary gland, ER-positive cells were localized in well-differentiated acinar cells and the epithelial cells of ducts. Although ER-positive cells were found the proliferative epithelial cells in adenosis and papillomatosis, the epithelial cells with apocrine metaplasia and/or cyst formation were ER-negative. Of 39 fibroadenoma cases, 11 cases were ER-positive. ER-positive cells were observed in their epithelial cells, and not in their interstitial cells. In three benign cystosarcoma phylloides cases, the epithelial cells were strongly ER-positive, while the interstitial cells were ER-negative. Of 122 breast cancer cases, 77 cases were ER-positive (63.1%). ER-positive cases were found in 61.4% in premenopausal status and in 65.4% in postmenopausal status without any significant difference between them. The ER-positive rate became lower in proportion to the increase in the tumor size, because of the increase in heterogeneous populations of ER-positive and ER-negative cancer cells ; the so-called mosaic pattern. And also the ER-positive rate was higher in the papillotubular carcinomas. In intracellular localization, cytoplasm, nuclei or both were occasionally stained, and intranuclear localization was supposed to correspond to localization of nuclear receptor.
