Studies of Colour Modification in Floral Tissues of Mogami-Benibana, a Cultivar of Carthamus tinctorius L.

The flower colour modification of Mogami-Benibana, a cultivar of Carthamus tinctorius L., shifts toward a bathochromatic side at pre-blooming or post-blooming. The colour shift is visualized and intensified by the accumulation of carthamin, which is derived from a yellow precursor named precarthamin through oxidative reaction by a carthamin-synthesizing enzyme. The red pigment was extracted from pistil and petal, which were collected by dissecting the flowers. It was then confirmed by TLC, HPLC, UV/VIS spectroscopic and spot scanning methods. The experimental data show that the red pigment are almost identical with an authentic carthamin. To investigate the effect of buffers on the carthamin formation studies were carried out in pistillary tissues. Three different buffers (50 mM) : the citrate-phosphate buffer, the citrate buffer and the acetate buffer, were used at varied pH values : 5.5,4.8 and 4.1. Acetate buffer (50 mM, pH 4.8) was found to be most promising on the red colouration. Ovary, petal, pistil and stamen were collected by dissecting carefully the flowering heads. They were laid separately on a slide glass and added a drop of Acetate buffer (50 mM, pH 4.8). Their reddening capacities investigated by using a flying spot scanner was as follows; Pistil>Petal>Stamen>Ovary. Pistillary tissue was divided into three parts : upper, middle and lower parts. The red colour appearance on each part was assayed by a technique of spot scanning. A potent activity was found in middle part. The reddening of upper part ranked next. The observed differences in red colour accumulation capacities are discussed briefly in relation to precursor translocation and enzyme activity distribution.
