The nephrotoxic effects of cis-Diamminedichloroplatinum (CDDP) were examined at 3 and 6h and at 1,and 3 and 10d in male rats at the ages of 52 weeks (middle age group ; MA group) and 9 weeks (young group ; Y group). CDDP was injected into rats in each group at the dose of 1mg/kg (i. v.) body weight. In the MA group, the levels of blood urea nitrogen and plasma creatinine were significantly increased at 3d, and the plasma K level was significantly decreased at 3d after injection. These changes were not seen in the Y group. From 6h to 3d after injection, the renal Pt level in the MA group was higher than that in the Y group. In the MA group, the levels of Zn, Cu, and Mn were significantly decreased from 1d to 10d after injection, the level of renal Ca was significantly increased at 6h, and the Mg level was significantly decreased at 3d after injection. There were no significant changes in the renal lipid peroxide level in either group. In the MA group, but not in the Y group, the superoxide dismutase activity was significantly decreased at 1d after injection. The level of renal metallothionein in the Y group increased beginning at 3d after injection, while that in the MA group decreased remarkably. Histomorphological examination demonstrated degeneration of the proximal tubuli in the MA group at 3d after injection. These results suggest that the renal disorders observed after the CDDP injection in the MA group were caused by mechanisms such as decreased cellular reaction associated with oxidative stress related to aging.
