The effects of repetitive stimulation on frog sciatic nerve fibers were examined. Supramaximal pulses of 1 to 100 Hz were used for stimulation for 60 min. Conduction velocity decreased gradually with the increase in stimulus frequencies ; a 60 % decrease from the control value, which was obtained by a single stimulation, was observed at 100 Hz stimulation for 60 min. The potassium concentration of the isolated single nerve fiber was examined with X-ray microanalyzer. The correlation coefficient of the potassium/phosphate ratio vs axonal diameter increased at stimulus frequencies higher than 10 Hz. These results suggested that repetitive stimulation caused an increase in potassium efflux from the intracellular space, and produced an accumulation of potassium ions at the circumferential extracellular space, Schwann cells and mesoaxons. The increase in the extracellular potassium concentration appears to decrease the membrane irritability of nerve fibers.
