酸性糖鎖の構造解析のための多次元HPLCマップ法

高橋禮子博士らによって開発されてきた多次元 HPLC マップ法は,今や中性糖鎖およびシアリル化糖鎖の構造解析において極めて有用な手法として広く認知されてきている。近年,硫酸化糖鎖,グルクロン酸含有糖鎖などの酸性糖鎖が細胞間のコミュニケーションを司っていると報告されていることから,HPLC マップ法の適用範囲をこうした酸性糖鎖まで拡張することにより,その有用性は一層の広がりをみせるものと期待される。そこで我々はこれまでに,硫酸基転移酵素およびグルクロン酸転移酵素を利用して,これら酸性糖鎖の HPLC マップを構築してきた。本稿では,酸性糖鎖の HPLC マップの開発に関して説明し,さらには本マップの応用例として硫酸基転移酵素 N-acetylglucosamine-6-O-sulfotransferase-1(GlcNAc6ST-1)の基質特異性の解析やインフルエンザウイルの糖鎖プロファイリングをとりあげ,HPLC マップの有用性について述べる。
